Samples are accepted at both extracted protein stage as well as at tissue or cell level. We have several validated extraction protocols which are used when samples are
submitted at tissue/cell level. Separation is typically performed using two dimensional liquid chromatography with a nanoflow HPLC system as second dimension but 1 dimensional analysis is available
on request. Samples should be submitted in clear, sterile tubes and must be clearly labelled with a printed label or indelible ink. The protein concentration must be indicated. Prior to sending
samples a client questionnaire and sample submission form must be completed and returned to the CPGR.
PMF - Samples for PMF analysis should be pure, gel resolved protein stained with either Coomasie or a MALDI compatible silver stain. The gel pieces should be cut into to 1mmX1mmX1mm and
dehydrated with acetonitrile before being desiccated in a vacuum centrifuge. The dry samples can be shipped to the CPGR at room temperature. Samples for in solution digest must be submitted in a
digest compatible buffer in clearly labelled tubes indicating ID and protein concentration. Pure proteins submitted in should be shipped at 4 C. Prior to sending samples a client questionnaire must
be completed and returned to the CPGR.
iTraq/ LCMS - Tissue/cells for extraction must be supplied frozen at -80C . The weight of the sample should be recorded before the samples are frozen. Samples already extracted must be
supplied in an iTRAQ compatible buffer (100mM triethylammonium bicarbonate) with excipients at trypsin compatible levels. If the sample is intended for iTRAQ ,a suitable, exogenous spike should be
included in the sample prep to control for sample loss during extraction. The protein concentration must be supplied with the sample. If the sample is supplied otherwise a sample prep fee may be
charged.
Mid Mass, High Mass - Protein samples for mid mass (m/z = 4000 -20 000) and high mass (20 000 -200 000) must be presented in analytical grade water free of salts or other
excipients.
